摘要: 本研究应用聚合酶链式反应(PCR),对现场捕获的携带肾综合征出血热(HFRS)病毒的动物肺组织,进行了病毒分离前后的分型扩增及扩增产物的限制性内切酶酶切分析比较,并与单克隆抗体(McAb)间接荧光法分型结果作了比较。初步表明,该方法可直接应用于现场捕获的动物肺所携带的HFRS病毒的血清分型研究。用PCR分型方法对不同宿主动物所携带的HFRS病毒的扩增特性还表明,黑龙江省黑河地区大林姬鼠和狗所携带的HFRS病毒,与黑线姬鼠所携带的HFRS病毒,可能存在较大的差异。

Abstract: This study compared the results of serotyping of lung tissues from HFRS virus-carrying animals captured in fields with PCR before and after virus isolation,those of analysis of digestion of amplified products with restriction endonuclease, and those of typing by indirect immunofluorescence with monoclonal antibody.Preliminary results showed that this method might be applied directly in serotyping studies.The characteristics of amplified HFRS virus carried by different reservoir animals studied with PCR typing showed that a rather big difference possibly existed between HFRS carried by Apodemus peninsulae and dog and that carried by Apodemus agrarius in Heihe District of Heilongjiang Province.
