小家鼠线粒体DNA控制区多态性和系统进化研究

Genetic polymorphisms and phylogenetic relationships of Mus musculus based on sequences of mitochondrial DNA D-loop

摘要: 目的 研究小家鼠线粒体基因组控制区(D-loop)的遗传多态性,为国境口岸鼠类鉴定和溯源提供技术支持。方法 以2011-2015年江苏口岸地区捕获的本底和输入性小家鼠为研究对象,设计引物扩增线粒体D-loop基因,对扩增产物进行测序后获得D-loop基因序列,进行基因多态性分析和遗传进化研究。结果 共获得21条国内外小家鼠D-loop基因序列,共有17个单倍型,测出533个序列位点,其中保守位点488个,可变异位点41个,核苷酸平均差异数为14.752 38。对21只小家鼠构建遗传进化树,分为3个分支。结论 小家鼠种群具有丰富的种内差异,其遗传进化规律与地理分布存在显著的相关性。

Abstract: Objective To understand the polymorphisms of D-loop gene from Mus musculus collected in Jiangsu ports, and provide support for the species identification of rodents from frontier port. Methods The D-loop genes from M. musculus were amplified, sequenced, aligned, analyzed genetic polymorphism and created genetic evolutionary tree. Results Twenty-one D-loop gene sequences were obtained from M. musculus, all of M. musculus shared 17 haploid type, 533 loci were measured in every sequence. There were 488 conservative loci and 41 mutation loci, the nucleotide differences number was 14.752 38 on average. The genetic evolutionary tree was created for 21 M. musculus, the genetic evolutionary tree can be divided into three branches. Conclusion Nucleotide polymorphism from M. musculus is rich. The high genetic diversity was indicated within M. musculus, which had a connection with geographical distribution.
