中国部分地区赫坎按蚊种团的分子鉴别研究

Molecular taxonomy of Anopheles hyrcanus group in some regions of China

摘要: 目的 阐明我国各地赫坎按蚊种团成员种的组成.方法 依据形态鉴定赫坎按蚊种团成员,种类应用PCR法和分子特征确认,分析的分子特征为rDNA-ITS2和rDNA-28S-D3序列.结果 对采自我国12个省20个地点的按蚊样本(n=1259)进行分子鉴别结果显示,该研究样本中包括赫坎按蚊种团成员种(n=1237)和其它按蚊3种(n=22);其中赫坎按蚊种团的成员种为中华按蚊、雷氏按蚊、八代按蚊、贵阳按蚊、筠连按蚊、克莱按蚊、比伦按蚊,以及3个未定名种(LL1、LL2和LL3);中华按蚊在大多数采集点密度最大.结论 赫坎按蚊种团非常复杂,综合分析确定其成员种类和分布非常重要.

Abstract: Objective To clarify the members of Anopheles hyrcanus group in some regions of China. Methods The members of An. hyrcanus group were identified by morphology, and the sibling species of An. hyrcanus group were determined by PCR and molecular identification. The molecular identification was based on the analysis of rDNA-ITS2 and rDNA-28S-D3 sequences. Results A total of 1259 Anopheles samples were collected from 20 sites in 12 provinces of China. The molecular identification results showed that 1237 of the samples belonged to An. hyrcanus group, and the remaining 22 belonged to 3 other species of Anopheles. The members of An. hyrcanus group were An. sinensis, An. lesteri, An. yatsushiroensis, An. kweiyangensis, An. junlianensis, An. kleini, An. belenrae, and 3 unidentified species (LL1, LL2, and LL3). The population density of An. sinensis was the highest in most collection sites. Conclusion The An. hyrcanus group is complex. It is important to identify the members of this group and their distribution by comprehensive analysis.
