Vero-E6细胞培养中汉坦病毒抗原流式细胞术检测方法的建立及与ELISA、IFA方法的比较

Establishment of a flow cytometry method detecting Hantavirus in Vero-E6 cells and its comparison with ELISA and IFA

摘要: 目的 建立运用流式细胞仪检测Vero-E6细胞中汉坦病毒抗原的方法,并与间接免疫荧光法(IFA)、ELISA进行比较。方法 将汉坦病毒感染10d的Vero-E6细胞以A35单克隆抗体为一抗,FITC标记羊抗鼠抗体为二抗,采用流式细胞仪检测表达抗原的细胞,并将病毒不同感染天数及病毒不同载量感染情况下的抗原检测结果与IFA、双抗体夹心法ELISA进行比较。结果 流式细胞术在感染后的第2天可检出的阳性细胞数为2.60%,其最低检出的病毒感染载量为50pfu/ml;而IFA和双抗体夹心法ELISA则分别在感染后的第3天、第2天能检出抗原,最低检出的病毒感染载量均为50pfu/ml。结论 流式细胞术检测法作为一种检测细胞内汉坦病毒抗原的方法具有简便、灵敏快速,与ELISA、IFA方法比较,可更直接准确定量。

Abstract: Objective To establish a flow cytometry method(FCM) to detect and quantitate the hantavirus(76-118) after amplification in Vero-E6 cell culture. Methods According to the developed assay,three methods detecting Hantavirus antigen including flow cytometry, ELISA and IFA were compared. The kinetics of the virus infection process was determined. Results The FCM could detect 2.60% of infected cells on the second days after inoculation, and the detected virus concentration was as low as 50 pfu/ml. ELISA caud detect antigen on the 2nd day after inoculation, however IFA caud detect antigen on the 3rd day. The virus concentration detected by both methods were as low as 50 pfu/ml. Conclusion Flow cytometry is a rapid,sensitive and accurate method to detect Hantaan virus antigen in Vero-E6 culture.
