摘要: 目的 检测共生菌沃尔巴克氏体(Wolbachia)在德国小蠊体内的感染情况。方法 应用沃尔巴克氏体wsp基因的1对通用引物(81F,691R),通过聚合酶链反应、T-A克隆和序列测定,将所得wsp基因序列与GenBank中的已知Wolbachia品系wsp基因进行同源性分析。结果 成功地从德国小蠊成虫的总DNA中扩增到一段317bp的wsp基因片段,与GenBank中的已知Wolbachia品系wsp基因同源性最高可达95%。结论 沃尔巴克氏体Wolbachia共生菌可能广泛存在于德国小蠊体内,为进一步研究德国小蠊的孤雌生殖与Wolbachia的相关关系等研究奠定了基础。

Abstract: Objective To detect and analyze wsp gene of Wolbachia symbiotic in Blattella germanica. Methods The fragments of the wsp gene of B.germanica was amplified by PCR with general wsp primers(81F, 691R). Target PCR product was purified, T-A cloned, sequenced and identified. The amplified fragment sequence was aligned with another published wsp genes. Results The presence of Wolbachia had been successfully detected in B.germanica, and the length of this gene is 317 bp. Multiple sequence alignment indicated that .germanica wsp gene was very similar to another eukaryotic gene in GenBank and shared more than 95% identity. Conclusion It shows that perhaps Wolbachia are bacteria widespread in B.germanica, which may lay foundation on the further study of correlativity between parthenogenesis of B.germanica and Molbachia.
