德国小蠊抗性与敏感品系细胞色素P450生化特征的研究

摘要: 目的对德国小蠊抗性与敏感品系细胞色素P450(Cyt-P450)的生化特征进行研究,摸索有关CytP450的研究方法,为进一步研究德国小蠊Cyt-P450与抗药性的关系打下基础。方法参照Omura和Sato的方法测定Cyt-P450含量,并记录差光谱。结果德国小蠊不同品系间Cyt-P450 CO差光谱的研究显示,敏感品系在450nm处有一明显特征峰,并在420nm附近呈现一失活峰,而抗性种群450nm吸收峰受到强烈抑制,420nm附近失活峰明显。结论德国小蠊抗性品系、敏感品系在Cyt-P450 CO差光谱上的差异,表明德国小蠊抗性品系的Cyt-P450参与了抗性的形成

Abstract: Objective To preliminary identify biochemical mechanism of insecticide resistance by studying CO difference spectra and content of cytochrome P450 in susceptible and resistant populations of German cockroach,Blattella germanica.Methods Activity and CO difference spectra of cytochrome P450 were made by method of Omura and Sato.Results The CO difference spectrum of Cyt P450 in susceptible strain showed maximal absorbance at 450 nm,but the absorption maximum was not observed at 450 nm in resistance population and the absorption in 420 nm was in evidence.Conclusion The results indicated that cytochrome P450 was correlated with resistance to insecticide in resistant strains of German cockroach.
