Abstract: Objective To study the colonial morphology of Yersinia pestis carrying bacteriophages and to provide a scientific basis for the accurate and rapid identification of plague pathogens. Methods Yersinia pestis carrying bacteriophages were cultured by Hottinger's agar streak method and/or antiphage serum agar method for colonial and cellular morphology observation with the naked eye and light microscope. Results Both samples contained typical Y. pestis as well as atypical Y. pestis with bacteriophages which had smaller colonies than typical strains due to the presence of bacteriophages and slow or inhibited cell growth. Conclusion Antiphage serum agar method and plate streak method allow identification of the colonial morphology of typical and atypical Y. pestis under a light microscope. These methods are simple, rapid, straightforward, and economical, and are of great significance for the identification of intractable pathogens of plague.
