免疫刺激剂对湖北钉螺酚氧化酶活性的影响

Effects of immunostimulants on phenoloxidase activity in Oncomelania hupensis

摘要: 目的 实验研究几种常见的免疫抑制剂与激动剂对湖北钉螺酚氧化酶活性的影响。方法 将实验钉螺置于4组免疫刺激剂植酸、维生素C、β-葡聚糖、脂多糖及去氯水对照组中浸泡60 min, 去壳, 取软体组织通过匀浆、离心分别获取粗制酶液, 以邻苯二酚为底物用比色法测定各组酶活力, 结果进行统计学分析。结果 对照组各时间段与0 h间比较, 差异均无统计学意义(F=2.10, P>0.05);植酸和维生素浸泡后各时间段与对照组比较酚氧化酶(PO)活力下降(P<0.05), 随着时间的延长活力逐渐下降, β-葡聚糖和脂多糖处理后PO活力上升(P<0.05), 且随着时间的延长PO活力为逐渐上升。结论 免疫刺激剂植酸、维生素C会引起钉螺PO活力下降, 而β-葡聚糖和脂多糖使PO升高。

Abstract: Objective To investigate the effects of several regular immunostimulants on the phenoloxidase (PO) activity in Oncomelania hupensis. Methods The snails were divided into four treatment groups and one control group. In the treatment groups, the snails were soaked in phytic acid, vitamin C, β-glucan, or lipopolysaccharid for 60 min, while in the control group, the snails were soaked in dechlorinated water for 60 min. After the shells were removed, the soft tissues of the snails were homogenized and centrifugated to obtain crude enzyme liquid. The PO activity in each group was measured by colorimetry with catechol as the substrate, and statistical analysis was performed on the measurement results. Results The PO activity at different time points showed no significant difference from that at 0 h in the control group (F=2.10, P>0.05). Compared with the control group, the phytic acid and vitamin C treatment groups showed significantly decreased PO activity at different time points after being soaked in phytic acid and vitamin C (P <0.05), and the PO activity decreased over time; the β-glucan and lipopolysaccharid treatment groups showed significantly increased PO activity after treatment (P<0.05), and the PO activity increased over time. Conclusion As immunostimulants, phytic acid and vitamin C can decrease the PO activity in O. hupensis, while β-glucan and 1ipopolysaccharid can increase the PO activity.
