从革螨和恙螨中检测到发热伴血小板减少综合征病毒核酸

Vector research of severe fever with thrombocytopenia syndrome virusin gamasid mites and chigger mites

摘要: 目的 调查革螨、恙螨携带发热伴血小板减少综合征布尼亚病毒(SFTSV)情况。方法 收集黑线姬鼠和山羊体表革螨和恙螨,按属种分类,用荧光定量PCR和常规RT-PCR法检测革螨和恙螨体内是否携带SFTSV基因组核酸。结果 黑线姬鼠体表收集的5组小盾纤恙螨SFTSV基因组核酸均为阳性,黑线姬鼠体表收集的3组毒棘厉螨和山羊体表收集的1组毒棘厉螨SFTSV基因组核酸均为阳性。基因测序结果显示,与SFTSVJS4株NP基因比对,其同源性在99%~100%。结论 小盾纤恙螨和毒棘厉螨均可携带SFTSV,具有传播SFTS的可能。

Abstract: Objective To examine whether gamasid mites or chigger mites carry severe fever with thrombocytopenia syndrome bunyavirus. Methods Gamasid mites and chigger mites were collected from Apodemus agrarius and goats body surface and the severe fever with thrombocytopenia syndrome virus(SFTSV)genomic nucleic acids were detected using fluorescent quantitative RT-PCR assay and RT-PCR. Results The five groups of Leptotrombidium scutellare collected from Apodemus agrarius showed positive results in SFTSV genomic nucleic acids. The Laelaps echidninus collected from A. agrarius and goats all showed positive results in SFTSV genomic nucleic acids. Gene sequencing results showed that the NP gene amplificated from positive samples had a homology between 99%-100% compared with that of the JS4 strain. Conclusion L. scutellare and L. echidninus are carrier of SFTSV and they may spread SFTS
