家蝇抗菌物质针刺诱导研究

摘要: 目的 探讨针刺体壁对家蝇抗菌物质诱导表达的规律。方法 室内饲养的家蝇Ⅲ龄幼虫,针刺体壁处理后,分别于第0、24、48、72、96h记录幼虫存活率,并提取其血淋巴,以金黄色葡萄球菌和溶壁微球菌作指示菌,采用平板法做抑菌试验,37℃恒温24h观察抑菌圈,并以抑菌圈直径表示抗菌活力的大小,借以指示家蝇抗菌物质的诱导产生规律。结果 经针刺诱导后,家蝇幼虫存活率逐渐降低,提取的血淋巴对金黄色葡萄球菌和溶壁微球菌的抑菌活力峰均出现在诱导后48h。结论 针刺体壁可诱导家蝇产生抗菌物质,血淋巴收集在诱导后48h最为合适。

Abstract: Objective To investigate the kinetics of the inducible antibacterial activity of the hemolymph in larvae of the housefly.Methods The third instar larvae were induced by pricking,living rates of the pricked larvae were recorded at different times after treatment,and hemolymphs collected at 0,24,48,72 hours were tested against Staphylococcus aureus and Micrococcus lysodeikticus respectively.The diameters of clear inhibitory ring were observed and recorded to express antibacterial activity.Results The living rates of the larvae declined gradually.The antibacterial activity peak appeared across 48 hours after treatment.Conclusion It implies that the antibacterial substance was induced in the housefly after being pricked.The immunized hemolymph should be collected at 48 hour after treatment.
