白细胞介素12对不同龄期大鼠尘螨性哮喘细胞因子的影响

摘要: 目的研究白细胞介素12对不同龄期大鼠尘螨性哮喘细胞因子的表达影响。方法取实验室人工培养尘螨,制备尘螨浸液,建立老年和成年大鼠尘螨性哮喘模型,用试剂盒测定白细胞介素12对哮喘大鼠细胞因子表达作用的影响。结果经白细胞介素12治疗后,老年哮喘大鼠白细胞介素5为(70±18)ng/L,淋巴细胞活化标志(sIL2R)为(0.60±0.40)mg/L,γ干扰素mRNA为(0.099±0.001)ng/L,白细胞介素4mRNA为(0.101±0.001)ng/L;成年致敏哮喘大鼠白细胞介素5为(53±20)ng/L,淋巴细胞活化标志(sIL2R)为(0.50±0.10)mg/L,γ干扰素mRNA为(0.109±0.001)ng/L,白细胞介素4mRNA为(0.094±0.002)ng/L。结论白细胞介素12对老年和成年大鼠尘螨性哮喘的影响与白细胞介素5、淋巴细胞活化标志(sIL2R)、γ干扰素mRNA、白细胞介素4mRNA比较差异有显著性(P<0.01)。

Abstract: Objective To study effect of Interleukin-12(IL-12) on cellular factors in dust mite asthmatics rats.Methods Dermatophagoides pteronyssinus extract produced asthmatics in rats of different ages.The cellular factors were determined by immune method.Results IL-12 inhibits antigen-induced airway hyperresponsiveness in asthmatics rats of different age.The old asthmatic rats:IL-5(70±18) ng/L,sIL-2R(0.60±0.40)mg/L,IFN-γ mRNA(0.099±0.001)ng/L,IL-4 mRNA(0.101±0.001)ng/L;The adult asthmatic rats:IL-5(53±20) ng/L,sIL-2R(0.50±0.10)mg/L,IFN-γ mRNA(0.109±0.001)ng/L,IL-4 mRNA(0.094±0.002)ng/L(P<0.01).Conclusion IL-12 remarkably effected on cellular factors in dust mite asthmatics of adult and old rats.
