大肠埃希菌O157∶H7蜚蠊实验带菌消长的研究

摘要: 目的:采用EHECO157∶H7-EDL933株,体外染菌感染德国小蠊(Blattela germanica),观察其体内外感染和带菌消长情况。方法:点滴法体外定量染菌,剂量为1μl(菌悬液浓度为8.0×105~1.9×107(CFU/ml),并采用昆虫单虫单管方法饲养观察。结果:蜚蠊经体外染菌,获得体内外带菌,体外带菌时间中位数为(144.0±22.17)h(95%CI:100.55~187.45h);体内中位数为(144±25.77)h(95%CI:93.48~194.52h);体内外带菌消长差异,无统计学意义(χ2=1.46,P=0.2263)。德国小蠊最长带菌时间至少216h(9d)。结论:蜚蠊不仅是EHEC-O157;H7的贮存宿主和疾病传播的媒介,更重要的是潜在的人类感染的传染源。

Abstract: Objective:The persistence of experimental Escherichia coli O157∶H7-EDL933 strain in cockroach(Blattela germanica) was studied.Methods:Cockroaches were externally contaminated by droplet 1 μl of EDL933 strain 8.0×105-1.9×107 CFU/ml,after contaminating O157∶H7,each insect was individually placed into a sterilized glass tube(15 mm×150mm) for 8 h-216 h.Results:The external and internal of cockroaches were infected with O157∶H7 by contaminating the pathogen.The median of external carriage O157∶H7 was 144.0 h±22.77h(95CI:100.55 h-187.45h);the median of internal was 144.0 h±22.77 h(95CI:93.48 h-194.52h).The difference of persistence curve of Escherichia coli O157∶H7 carriage in external and internal of cockroaches was no statistically significance through Log Rank test(χ2=1.46,P=0.2263).Droplet experiments of O157∶H7 to cockroaches showed that the ingested bacteria were harbored in the intestine of cockroaches and continued to be excreted at least for 216 h(9 d).Conclusion:The result showed that the cockroaches were not simple reservoirs and mechanical vectors of EHEC O157∶H7,but an epidemiological potential source of O157∶H7 infection for humans.
