微板法检测蚊虫体内乙酰胆碱酯酶活性研究

摘要: 目的:检测不同敏感品系(S)蚊虫和室内选育的抗性品系淡色库蚊体内乙酰胆碱酯酶(AchE)活性。方法:微滴定板法。结果:4种S品系蚊虫中,以中华按蚊体内AchE水平最高;3种抗性品系蚊虫中,抗残杀威品系(Rp)的AchE活性>抗氯氰菊酯品系(Rc)>抗DDVP品系(Rd),且后者与S品系差异无显著性。蚊虫体内AchE活性随发育而逐步增高。以不同浓度残杀威作抑制剂,测定室内不同品系淡色库蚊个体的AchE活性,表明蚊虫AchE活性的抑制率随抑制浓度的提高而增加,呈正比关系;在0.01mmol/L浓度残杀威的抑制下,AchE的抑制率S品系>Rc品系>Rd品系>Rp品系。结论:3种抗性品系的AchE的敏感性均低于S品系。

Abstract: Microplate test was employed for the detection of acetylcholinesterase(AchE) level in susceptible mosquitoes and resistant strains of Culex pipiens pallens.Among 4 species of susceptible mosquitoes,Anopheles sinensis produced the highest level of AchE. The AchE level varied in different resistant strains of Cx. pipiens pallens, with anti propoxur strain the highest level,foollwed by anti-cypermethrin strain and with anti-DDVP strain the lowest. The AchE activity increased with the development of mosquito. Using propoxur(at 0.01mmol/L) as the inhibitor , the inhibition rate of AchE was highest in susceptible strain,followed by anti-cypermethrin strain ,anti-DDVP strain and anti-propoxur strain ,in descending order.The sensitivity of AchE was higher in susceptible strain than in resistant strains.
