Typing of Hantavirus from Lung Tissues of R. norvegicus by Reverse Transcription-Polymerase Chain Reaction and Sequence Analysis

摘要: 对来自北京西客站附近一集装箱仓库的2份汉坦病毒免疫荧光抗原阳性褐家鼠肺组织进行分型研究。方法;采用逆转录聚合酶链反应(RT.PCR)和序列分析。结果:这两份褐家鼠肺标本均携带汉城型(SEO)汉坦病毒(HV)。RT-PCR扩增其中1份标本所携带病毒的M基因330bp片段,通过pGEM-T载体克隆进入大肠杆菌JM109,并进行核着酸序列测定,经与其它HV核着酸序列的同源性比较,其与SEO型毒株相应片段核着酸序列的同源性在93.9%~96.4%之间,而与其它血清型毒株的同源性在61.5%~75.8%之间。结论:首次证实北京地区宿主动物携带SEO型HV.

Abstract: The purpose of thls study was to type 2 strains of hantavirus frorn antigen positive lung tissues of R.norvegicus captured in a container Pool near Beijing railway station.Reverse transcriptionpolyrnerase chain reaction (RT-PCR) showed that the 2 samples carried hantaviruses of SEO serotype.330 base pair region of the medium (M) genome segment of hantavirus was amplified directly frorn one of the antigen positive rat lungs by RT-PCR and cloned into E.coli JM109 by using plasmid vector pGEM-T.The genomic fragment was sequenced and compared with those of other well characterized hantaviruses.The comparison of the sarnple to SEO virus isolates revealed 93.9% to 96.4% homology and displayed 61.5% to 75.8% homology when compared with other serotypes of hantavirus.The existence of SEO virus in host animals in Beijing was verified for the first time.
