淡色库蚊细胞早熟凝聚染色体的诱导

摘要: 用淡色库蚊建立的细胞系(Cpp512)细胞与中国仓鼠卵巢细胞融合,诱导出蚊细胞的早熟凝聚体(PCC)。融合剂使用分子量为4000的50%聚二乙醇(PEG),有丝分裂细胞与间期细胞比例为1;15,在32℃下融合2~3min,温育时间为1h。早熟凝聚染色体分三种基本类型。G1-PCC为单股,早期比晚期浓缩;S-PCC呈“粉碎”样外观;G2-PCC为双股,分散好,数目可数,类似于有丝分裂期染色体,但更长,故可用作高分辨显带及其它细胞遗传学的进一步研究。PCC技术可成为实用有效的蚊细胞遗传学研究技术。

Abstract: Premature condensed chromosome (PCC) of mosquito cells (Cpp512 cell line, established from Culex pipiens pallans)was induced by fusing with mitotic Chinese Hamster Ovary(CHO) cells, using 5)% polyethylene glycol(PEG, MW=4000)as fusogen. The ratio of the mitotic cells to interphase cells was 1: 15. The fusion was performed at 32℃ for 2-3 min., then incubated for 1 h. The PCC morphology clearly showed 3 kinds of essential types: G1-PCC had single chromatid, the early phase was more condensed than later one: S-PCC had a "pulverized" appearance; G2-PCC had had donble chromatids which were well spread and countable, just like the mitotic chromosome but much longer than the mitotic one, G2-PCC therefore could be used for high resolution chromosome studies. The PCC technique would be useful and powerful tools in mosquito cytogenetic study.
