吉林省延边朝鲜族自治州东瀛伊蚊的发现与鉴定

Discovery and identification of Aedes nipponicus in Yanbian Korean Autonomous Prefecture, Jilin Province, China

摘要: 目的 对吉林省延边朝鲜族自治州(延边州)采获的存疑伊蚊标本进行物种鉴定,记录本地蚊虫种类。方法 对2024年 8月在延边州龙井市森林中人工诱捕采获的存疑伊蚊成虫标本进行形态学初步鉴定,用PCR扩增其线粒体细胞色素C氧化酶亚基Ⅰ(COⅠ)基因并测序,与GenBank中的序列进行比对,分析相似性,通过MEGA 6.0软件构建系统发育树。结果 采获标本中有5 只雌性伊蚊与纷蚊亚属(Finlaya)银雪伊蚊(Aedes alboniveus Barraud,1934)、东瀛伊蚊(Ae. nipponicus LaCasse et Yamaguti,1948)、新雪伊蚊(Ae. novoniveus Barraud,1934)具有相似的形态特征。随机选取其中4只蚊虫的COⅠ基因序列与GenBank中已知东瀛伊蚊序列比对,相似度均>99%。遗传进化分析显示,该蚊与已知东瀛伊蚊序列聚为同一分支。结论 东瀛伊蚊在延边州首次被发现,为当地蚊虫的新记录种,为东瀛伊蚊在吉林省的分布提供了明确证据。标本保存于延边州疾病预防控制中心。

Abstract: Objective To identify the species of suspected Aedes mosquito specimens collected from Yanbian Korean Autonomous Prefecture, Jilin Province (Yanbian Prefecture), and to record the local mosquito species. Methods Preliminary morphological identification was performed on suspected adult Aedes mosquitoes captured by human landing collection in the forest in Longjing, Yanbian Prefecture in August 2024. The mitochondrial cytochrome c oxidase subunit Ⅰ (COⅠ) gene was amplified using PCR, and sequencing was performed. The sequences were compared with those in GenBank for similarity, and a phylogenetic tree was constructed using MEGA 6.0 software. Results Five female Aedes mosquitoes were found to share morphological characteristics with Ae. alboniveus Barraud, 1934, Ae. nipponicus LaCasse et Yamaguti, 1948, and Ae. novoniveus Barraud, 1934 in the subgenus Finlaya. TheCOⅠ gene sequences of four randomly selected mosquitoes showed over 99% similarity to known sequences of Ae. nipponicus in GenBank. The phylogenetic tree analysis indicated that they clustered with known sequences of Ae. nipponicus. Conclusions Ae. nipponicus has been discovered in Yanbian Prefecture for the first time, representing a new record of mosquito species in the area. This also provides clear evidence of the distribution of Ae. nipponicus in Jilin Province. The specimens are preserved at the Yanbian Center for Disease Control and Prevention.
