Abstract: This paper reported the stability and high expression of plasmids of genetically engineered larvicidal Anabaena.The results showed that propagated and cultured in solid medium without antibiotic selection for 3-5 years,the three recombinant Anabaenas kept their resistance to antibiotic and their high larvicidal activity.Their LC50 (24h) to Culex pipiens pallens were 8.26×103 cells/ml (A.7120 pDC26)、2.62×104 cells/ml (A.subtropic pDC26) and 4.21×103 cells/ml (A.cylindrica tox) respectively.So the genetically engineered Anabaena might be a prospective insecticide for field control on mosquito and the shuttle plasmid might be beneficial to the expression of other genes.
摘要: 目的:探讨基因工程藻高效杀蚊幼活性,以确定其质粒表达情况。方法:抗性筛选及蚊虫毒效测试。结果:随机筛选出的基因工程单藻落无一抗性丢失,且3种基因工程藻对淡色库蚊仍具较高毒效,LC50(24小时)A.7120 pDC26为8.26×103cels/ml;A.SubtropicpDC26为2.62×104cels/ml;A.Cylindrica tox为4.21×103cels/ml。结论:穿梭质粒在基因工程藻细胞内稳定遗传,并持续高效表达。
