Optimization of fermentation conditions of endophyte JJ18 of Pseudolarix kaempferi Gord for molluscicidal effect

具有灭螺活性的金钱松内生真菌JJ18发酵条件优化

Abstract: Objective To improve the molluscicidal effect by optimization of fermentation medium of endophyte JJ18 from Pseudolarix kaempferi Gord. Methods The medium composition and the cultivation conditions were optimized by orthogonal and single factor experiment. The immersion method suggested by WHO was applied to determine the effect and the main assessment index was mortality of the snails and biomass of fermantation liquid. Results This optimum culture medium included potato of 175 g/L, initial pH 6.0, glucose of 15 g/L, sucrose of 15 g/L, KH2PO4 of 0.5 mmol/L, the inoculum dose of 2%, the medium content of 50 ml, temperature of 30 ℃, incubation of 7 d and 120 r/min. The mortality of the snails was 100% with the optimum culture medium, which was increased by 16.7%. There exists a certain correlation between biomass of endophyte JJ18 and mortality. Conclusion The mortality is improved with optimization of fermentation techniques, so endophyte JJ18 of Pseudolarix kaempferi Gord has potential for molluscicidal application.

摘要: 目的 通过改进发酵工艺提高金钱松内生真菌JJ18的杀螺活性。方法 采用单因素和正交试验对发酵培养基及发酵条件进行优化。按照WHO的杀螺剂浸泡试验法,以杀螺率为主要衡量指标,同时考察发酵液的生物量,对金钱松内生真菌JJ18发酵条件进行优化。结果 以马铃薯175g/L为氮源,初始pH值6.0,葡萄糖和蔗糖15g/L为碳源,磷酸二氢钾0.5mmol/L为代谢调节因子,接种量2%,250ml三角瓶装液量50ml,温度30℃,120r/min,发酵7d,杀螺活性最大提高16.7%。生物量与杀螺率存在一定的相关性。结论 通过改变发酵工艺可以提高杀螺活性,金钱松内生真菌JJ18具有开发应用的潜力。
