滤纸法测定淡色库蚊非特异性酯酶和乙酰胆碱酯酶活性的实验研究

Abstract: Objective:To establish the filter paper test for the determination on non-specific esterase and acetylchotin esterase as a resistant determination method.Method:Filter paper test.Result:Filter paper test was used in this study to test the non-specific esterase(NSE)and acetylchotin(AchE)activities of susceptible and resistant strain of Cx.pipiens pallens,so as to determine the resistant level of individual mosquito according to colour changes.The results indicated that susceptible mosquitoes showed no or slight colour reaction while resistant mosquitoes revealed deep colour,and the NSE and AchE activity was inhabited in susceptible individuals by insecticides,while it cannot be inhabited in resistant individuals.It was demonstrated that there existed the difference in the AchE activity between susceptible and resistant mosquitoes.Conclusion:This method can rapidly determine the NSE and AchE activity in mosquitoes,and easy to perform,so it can be used for qualitatively determining the resistant level of mosquitoes and be convenient to be popularized in the field.

摘要: 目的:测定淡色库蚊敏感品系和抗性品系的非特异性酯酶(NSE)和乙酰胆碱酯酶(AchE)活性状况,区分单个蚊虫的抗性情况。方法:滤纸法。结果:敏感品系个体的NSE和AchE在滤纸上显色较浅或不显色,抗性品系个体则显色较深。采用不同浓度杀虫剂对蚊虫AchE活性进行抑制试验,敏感品系个体的AchE随杀虫剂浓度的增加而被抑制(不显色),抗性品系个体则不被抑制(显色明显)。结论:两品系间AchE的含量和活性有所不同。滤纸法可快速地测定淡色库蚊内的NSE和AchE活性,简单易行,可作为一种淡色库蚊抗药性测定方法,便于在现场中推广应用。
