Purification and characterization of the larvicidal toxin of Bacillus sphaericus strain C 3-41

The conditions of extraction and parification of the larvicidal toxins of Bacillus sphaericus C 3-41 and Bs -10 and the toxin characteristics were reported in this paper. Preliminary extract of toxic protein was obtained by freezing and thawing the spore-arystal complex. Then the larvicidal toxins were purified by Sephadex G-75 and DEAE-52 chromatography. When subjected to PAGE under denaturing conditions, the toxic proteins of strains C 3-41 and Bs -10 migrated in positions corresponding to 40 kilodaltons(kDa). The LC 50 values of the toxins of C 3-41 and Bs -10 against the third instar larvae of Culex quinquefasciatus were 26.99 and 37.92ng/ml respectively. The toxic proteins from strain C 3-41 and Bs -10 were not stable by heat-treatment for 20 minutes at 50-80℃ and by treatment with 0.05-0.2N NaOH for 24 hrs. The toxicity of the larvicidal toxins can be destroyed by pronase but not by trypsin. The experimental results showed that both hydrolysazed toxins by acid were composed of sixteen kinds of amino acid and the toxic composition of C 3-41 and Bs -10 were similar to those of B.s.2297 and Bacillus thuringiensis subspecies israelensis.
Vertical distribution of Aedes albopictus and Characteristics of the Community of Mosquitoes at Bamboo Forests in Naping

This paper report a survey of mosquito species at various altitude above sea-level in 5 bamboo forests in Naping Altogether there are s genera and 19 species of mosquitoes had been collected from August to september in 1986. Ae.albopictus was the dominant species. It occurred in 110m bamboo-holes examined with a frequency of 78.57%. But the population decrease associated with the altitude increase in the bamboo forests. It was not found where altitude are over 800m. From a sharp contrast, Ae. fengi was the dominant one at various altitude level except the zone 110m. The population increase company with the altitude increase. It was Caught with a frequency of 85.29% in 1020m. above sea-level.

According to the experimental results of acute oral toxicity (LD 50>5000mg/kg), acute dermal toxicity(LD 50>3000mg/kg), acute inhalation toxicity(LC 50>10g/M 3), and no irritation on conjunctiva of rabbit, the acute toxicity of methoxyphenyl acetata (8701) on animal belongs to the category of low toxicity. Ames's test was "negative". The results of the micronucleus test and the teratosperm test showed that there were no significance (P>0.05) in the test of significance between the treatment and its control. The teratological and mutagenic action of "8701" on animal was also not obserued. The result of subacute toxicity indicated that "8701" was no effect on the rate of growth and development of mouse. The accumulative toxicity of "8701" also belongs to weak accumulative drug (K>5.3).

In order to develop the application of Methoxyphenyl Acetate in pest control, it is important reference to clear the relation between the temperature and gravity of this compound. So the experiment was carried out in the Thermo-gravity instrument (Perkin-Elimer TGS-type 2). The result showed that there is a relationship between increasing the temperature and decreasing the weight of the compound during the temperature from 120℃—240℃. Recovery of the product content is more than 87% by GC analysis, and the GC-MS spectrum is shown that the compound was not decomposed after heating treatment.

125I isotope was used in marking the blood of the white mice Mus musculus to study the bloodsucking activity of Xenopsylla cheopis & Leptopsylla segnis, including reproducing adults and new emergence adults, at 20℃, 28℃ and 35℃ temperature conditions. The results are as follows: 1. The blood-sucking capacities of reproducing adults, for both of the two species of fleas, are greater than that of new emergence adults in 20℃, but no obvious defference in higher temperature. 2. The blood-sucking capacity of X. cheopis is greater in 35℃ than that in lower temperatur, but it is on the contrary for L.segnis. 3. Both of the two species of fleas appear no obvious difference in blood-sucking rates between two sexes of adults under same experimental conditions. 4. The blood-sucking speed of X.cheopis is much quicker than that of L. segnis. 5. The results also show that if the sucking time is long enough the young adults, both two species, possess blood-sucking ability as strong as the old adults, especially in higher temperature coditions.
Studies on Population Dynamics and Breeding Ecology of Rattus flavipectus in Putian Area

1. This wook was carried out in Putian county, from Mar 1987 to Feb. 1989. 230 specimens were collected. Average capture rate was 1.60%. being 8.10% of the whole house rodents. 2. There are obvious seasonal fluctuation of the rodential population density and percentage of specific composition. The peak of population density occunred in Dec. to Jan. 3. The sex ratio of Rattus flavipectus was 0.85 (♂/♀). The percentage of lower testis of males was 73.58%. The pregnancy rate was 22.58%. The average litter size was 6.18+0.2866. The reproductive index was 0.7522. There are seasonal and annual variations of reproductive parameters. 4. The population density and percentage of specific composition had been declining continually from 1950s to 1980s in Rattus flavipectus, and this present probably correlation with the earths chage into warm.
The Investigation on Hibernation & Emergence from hibernation of Fivetoed Jerboa

The hibernation and emergence from hibernation of five-toed jerboa were studied in Huhehot district. After further analysis, some results are as follows: After long hibernation, the five-toed jerboas emerge from hibernation at the last few days in March or at the first few days in April. The critical daily average temperature of emergence from hibernation is 3.3-4.2℃. The sequence of emergence from hibernation is that after the males have emerged about 20days, the females begin to emerge. The major members of emerged rodents are the adult 2-4 (Ⅳ~Ⅵ). There are no juveniles' emerging and only a few seniors and subadults emerge from hibernation. The beginning dates of hibernation are the last few days in September or the first few days in October. The critical daily average temperature of hibernation is about 14℃. The sequence of hibernation is that after the females have hibernated some days the males begin to hibernate. The finishing date of hibernation is on October 20th. There are no obvious sequence of hibernation aud emergence from hibernation in aged-groups.
Survey on distribution and seasonal variation of Suncus murinus (Insectivora)

It was reported that ecology, spatial distribution and seasonal variation of Suncus marinas has been suryeyed at the miltitary camp area. The results showed that the Insectivora mainly habited and move about in official room, kitchen and chichen-roost, and so on. The peak of seasonal variation is from September to Norvenber, and from December to next January is lower season for Insectivora. After then it is gradually raising in the population in March this will be the circle of every year. The ratio of Insectivora to domestic rodents would be infleance by rodenticite in domestic.
The investigation of Lyme disease in the southern part of the Greater Xing-An mountains

From April to June 1991, investigation of Lyme disease was carried out in the southern part of the Greater Xing-An mountains. The sera of 496 residents were tested for antibodies against Borrelia burgdorferi (strain B31) by indirect immnnofluorescence assay, 38(7.66%) residents were positive. 22 cases of Lyme disease were confirmed by clinical manifestation and serological test. 4 isolates were made from 26 Ixodes persulcatus, These isolates were distinguished between China and USA by monoclonal antibodies. These studies indicate that this region is endemic area of Lyme disease.

A high level of 6088-fold population (RPy strain) of fenvalerate resistant Culex pipiens pallens(Coq.)collected from Nanshi district in Shanghai was successfully developed in laboratory by treating the 4th instar larvae with fenvalerate. The Rpy strain showed high cross-resistant level for P, P'-DDT(>388X), for other Pyrethroids [6(Tetramethrin) to>140X (tran-permethrin)]. For Dieldrin, Dimethoate, Trichlorfon and Propoxur (1-2X), and for Fenitrothion, Methylparathion, Malathion, Temephos and Carbaryl (more susceptible than S strain).The in vivo studies of synergism by different special inhibitors such as Pb, TPP and DMC indicated that mechanisms of resistance in the Rpy strain might be associated with DDTase and mixed-function oxidase (MFO) for DDT-resistance, and MFO and Esterase for fenvalerateresistance.

The early 4th instar larvae of Culex pipiens pallens were treated with BS C 3-41 solution at a concentration of 0.01ppm in the laboratory. The life table characteristics of the F 1 generation of the mosquito were studied. In comparison with the untreat control group, the net reproduction rate of the treated mosquitoes reduced by 19.23%, while the average duration of the life cycles increased by 4 day; on the other hand, F 1 generation showed an effect of subsifing in the hatch rate. Jt is thus speculated that population of the F2 generation would be diminished due to the effects of BS C 3-41 on the previous generation.

Studies on the mosquito net treated with insecticide in long acting netimpregnating formula to control the mosquito invading into the experiment hut

With the luring cow we carried out a study on the deadly poisonous and excito-repellent effect of the mosquito net treated with insecticide in long acting net-impregnating formula on the mosquito invading into the specially constructed experimental hut in the surburb of Yangzhou City from June to September of 1990. From 1-80days after treatment in 80mg ai/m 2, the results showed that the treated mosquito net played an ideal role in the controlling of the local main species An. sinensis and Cx. tritaeniorhynchus, and the total mean mortalities during this period were 79.1% and 53.6% respectively. It was also revealed that the number of the mosquitoes invading into the treated hut reduced for 46.3% compared with that observed in control hut. The rate of mosquito flying out of the hut window was much higher in treated hat (76.4%)than that in the control hut (52.6%) (P<0.01), which proved that the long acting net-impregnaing formula has an excito-repellent effect on mosquitoes.

Deltamethrin 2.5% emulfowable (supplied by Roussel-Uclaf, France, under the trade name Dorine)was used at the dosage of 12.5mg ai/m to spray all houses of mosquitoes and flies rest places in the study area. The mosquitoes density was checked by Black Light-trap Box. The results shows that in term of RPI(Relative Popalation Index)as compared with pretreatment and control data the Calex quinquefascitatus mean density decreased by 74.9% after three months treatment and the Musca domestica decreased by 97% after four days treatment.

From 1989 to 1991 a large scale of integrated control program for cochroach were carried out in Yue-Yang area. The result shows that there are two domiaantnt species in the local, that is periplanets americana and Periplaneta fuliginosa. According to the pilot esperiment in laboratory and field, and geographical feature and weather condition the control program for cochroach was confirmed, namely, mainly to improve enviromental sanitation was chief manner, and associated with chemical and physical control against cochroach is sccond, and organizing mass movement control were in a large scale, advocate and promated widely to eliminating cochroach's Oothaca, and at the samtime, spraying insecticite—Deltamethrin poder and emulsion it courd be as soon as posibble to reduced the density of the insects and eliminated the infestation. Studies on the Infest Rate and Control of Cockroach in the Productive Workshops

The paper reported the result that studies on the infest rate and control of cockroach in 35 productive workshops in Shanghai. Two species of cockroach, Periplaneta emarginata and P. americana are discovered in the productive workshops. The result showed that the infest rate of cockroach were 58.85%. An average density of P. emarginata per square meter was 10.65 in 20 workshop, but P. americana was 38.50 only in 8 workshops. On the spot used 1% Acephate sugar liquid asan insecticide against cockroach were carried in the dry wokshops or basement, the average pesticidal rate was 95.89% for 24 hours after.
