摘要: 目的:研究鼠疫耶尔森氏菌的插入序列IS 1541的插入位点的特性。方法:用一条锚定的IS 1541内部锚定引物和一条随机引物进行PCR扩增;核酸分子杂交技术;克隆;测序。结果:14株鼠疫菌的扩增图谱基本相同,杂交图谱差异,可分成3种类型。克隆、测序其中一个片段,其序列仅在两端的引物区与IS 1541同源,而与E.coliK-12 MG 1655的 prlc 基因同源性较高。结论:IS 1541在鼠疫菌中的分布有差异,可以作为该菌的基因标志,用于分型。

Abstract: Objective: Studying on the character of IS 1541 of Yersinia pestis. Methods: PCR with an anchoring primer in IS 1541 and random primer; southern blot; cloning; sequence. Results: The PCR map of 14 strains were basically the same. The southern blot map were different. They were divided into three types. One fragment were selected to cloning and sequence. In its sequences, only primer region sequences on both ends were homology with IS 1541, but it was highly homology with prlc gene of E.coli K-12 MG 1655. Conclusion: The distribution of IS 1541 in Yersinia pestis was different. It can be a gene marker and used in gene typing
